FOXO4-DRI is a synthetic peptide built from D-amino acids in reversed (retro-inverso) sequence order. It is based on a region of the FOXO4 transcription factor that contacts p53. The D-retro-inverso design resists protease digestion in cell-culture conditions.
In published cell-culture work, FOXO4-DRI is used as a tool compound to study the FOXO4–p53 binding interface. Assays include co-immunoprecipitation, p53 nuclear localization imaging, and caspase-activation readouts in senescence-induced cell lines.
